Super Resolution Imaging using Fluorescence Spectroscopy

Python pipeline for single-molecule localization microscopy with pTIRF microscope SOP.

09/2025RGIPT, Jais, UP

Guided by: Dr. Debashis Panda | Associate Professor, Department of Chemistry, RGIPT

Utilized Python to develop a comprehensive software pipeline for high-precision single-molecule localization microscopy (SMLM) data, incorporating 2D Gaussian fitting, background subtraction, and multi-stage drift correction to produce super-resolved datasets. Authored a comprehensive standard operating procedure for a prism-type Total Internal Reflection Fluorescence (pTIRF) microscope.

Key Highlights

  • Python pipeline for SMLM data with 2D Gaussian fitting
  • Multi-stage drift correction for super-resolved datasets
  • Authored SOP for prism-type TIRF microscope
  • Data analysis using Origin and Olympus IX73 Microscope
